Research interests

Global trends in pharmaceutics suggest that biomacromolecules such as proteins and nucleic acids will be among the drugs of the future. This is, in part, because they can catalyze specific reactions, bind to targets, modulate protein expression and protein–protein interactions, edit genes, etc. These mechanisms of action are typically not available to small-molecule drugs, which provides interesting opportunities for treating ‘undruggable’ diseases. Moreover, these molecules can be engineered by editing their amino/nucleic acid sequence or by chemical modification to tune properties. This offers exciting opportunities for exploring structure–activity relationships, elucidating new biophysical phenomena, characterizing mechanisms of action, and developing new tools for biotechnology.

Over the last six years, my group has navigated this research space along three major research axes. Our first has consolidated a platform of ‘molecular sieving’ polymer coatings for immunogenic anti-cancer enzymes, to preserve activity while isolating them from the immune system. We have been the first to demonstrate that branched polymers grafted to proteins can yield discontinuous coatings with tunable nanoscale pores. We have extensively characterized the molecular sieving properties of these coatings in vitro, in vivo, and in silico, and have developed predictive tools for de novo coatings [Small Methods 2025, Mater Today Chem 2022 & 2019, from original work in Nature Commun 2014, Adv Funct Mater 2013].1-5 Our second established a modular DNA nanotechnology platform to rapidly make anti-cancer bispecific antibodies (bsAb). We developed the synthetic/analytical framework to prepare libraries of DNA nanostructures bearing multiple protein ligands at precise locations, and have tested these constructs as scavengers for toxic amyloid proteo-species and as bispecific T-cell engagers [Aggregate 2024, J Am Chem Soc 2024 & 2019].6-8 We have also used this platform in an ultrasensitive quantum biosensor [Nano Letters 2025].9 Our third, more general, deepened our knowledge of the biophysics of bioconjugates through a portfolio of projects, to search for new phenomena and elusive properties and thus remain avant-garde in the field. We have developed new reactions to reversibly conjugate polymers to proteins,10, 11 studied the mechanisms of protein inactivation caused by polymer grafting,5, 12 and explored new routes for modulating protein–protein and protein–polymer interactions in solution.13-15 In the wake of covid-19, we equally explored routes for inactivating viruses by stimulating long-range collective vibrations of their capsids using femtosecond lasers (collaborative research)16, 17 and developed self-sanitizing/self-cleaning coatings for frequently touched surfaces (and founded Nanosaber Coating Technologies Inc.).18 Finally, we developed a long-acting bioconjugate with neuroprotective properties, and I led the European consortium (project GOTTARG) to validate its mechanism of action in rats and non-human primates,19-22 showing promise for humans. Overall, we have invested strongly in pushing the boundaries of bioconjugate design, production, analytics, and control.
